Test for amphetamine in its infancy is based imunohromatograficheski test in vitro. It is designed to identify and kolchistveno deterministic amphetamines in human urine . The presence of amphetamine in human urine at the border level 1000 ng / mL can be detected for 3 minutes.

SUMMARY AND EXPLANATION
Amphetamines are a class of potent sympathominetic agents with therapeutic applications . The most common amphetamines are d- amphetamine and d, l- amphetamine . They are mainly stimulants of the central nervous system and cause the release of nervotransmitarite epinervina , noreshshervina and dopamine in the brain and body , leading to the euphoria , agitation with a sense of energy, self-confidence and mobility. Chronic abuse of amphetamine leads to tolerance and opioid effect. Cardiovascular responses to amphetamine include increased blood pressure and arrhythmia, yet it causes anxiety, anxiety , paranoia, hallucinations and psychotic behavior. Amphetamine is metabolized by many paths . In general, acid urine promotes excretion whereas alkaline vaprepyatstvuva it . In 24 hours approximately 79% of the amphetamine is released in acidic urine , and about 45% in the alkali . Characteristically and excretion of 20% as unchanged amphetamine . This amount can be detected within 1-2 days after use .
Amphetamine assay is based on the principle of specific immunochemical reaction between antibodies and antigens to be analyzed on the particular components in human urine specimen . The quantitative analysis is based on competition for antibody binding between the compound of the drug and free drug which may be present in the urine sample to be analyzed . When the urine sample is drug , it competes with the compound of drugs for the limited amount of antibody - dye compound . When the amount of drug is equal or more than 1,000 yl / mg , it will predovrati binding of free drug antibody . Therefore, a positive urine specimen will not show a colored band in the test area and will have a positive result , while the presence of a colored band indicates a negative result.
The test has a control line that serves as a procedural control . This colored band should always appear in the control line region if the test is kept in good condition and has been performed correctly .

MATERIALS PROVIDED
1. Instructions.
2 . Test kits Amphetamines

REQUIRED . BUT uninsured MATERIALS
1. Container for the collection of urine
2 . Timer or clock. STORAGE AND STABILITY The test kit should be stored at 2 to 30 ° C and is effective until the expiration date.

PRECAUTIONS
Only in vitro and judicial upotreba.Ne use the product after the expiration godnost.Rabotete carefully samples as may be zarazni.Ne open the foil pouch until you are ready to testa.Izpolzvayte new container for each urine sample for to avoid contamination of one sample by another sample

SPECIMEN COLLECTION AND PREPARATION
For each test sample required approximately 150mI fresh urine samples require no special handling or processing. The sample should be taken in a clean, dry , plastic or glass container. If the quantitative test is not carried out immediately , the urine sample can be cooled to 2-8 C and stored up to 7 days or frozen samples must reach room temperature before testing. Urine specimens that show a large precipitate or turbidity should be centrifuged or allowed to settle, so as to obtain a clear liquid for testing.

QUALITY CONTROL
Control band is an internal reagent and procedural control . It will appear if the test has been performed correctly and the reagents are reactivated . The control standard can be used to validate the performance of the reagents , and to determine the reliability of the test. The controls are not provided with this test can be purchased .

PROCEDURE
Test strip
1. Leave all materials and specimens to reach room temperature.
2 . Remove the test strip from the sealed foil.

3 . Dip the strip into the urine sample with the arrow pointing to it. The level of the sample must not be higher than the maximum indicated by the arrow .
4 . Hold the strip in the urine until a colored pink colored band (approximately 20 seconds) .
5 . Remove the tape and place it on a clean, non-absorbent surface, face up, or leave it in the urine, if the level of the sample is higher than the maximum indicated by the arrow .
6 . Read the result between 3-8 minutes.

For a deck of cards
1. Leave all materials and specimens to reach room temperature.
2 . Remove the test card from the sealed foil.
3 . Place the pipette in the specimen and squeeze it to get the sample
4 . Hold the pipette in a vertical position over the sample of the test card and put 2-3 drops of specimen ( 100-150 ?l)
5 . Read the result between 3-8 minutes.
Do not interpret results after 10 minutes.

INTERPRETATION OF RESULTS
Negative: Two colored bands. The appearance of two bands stained in a line in the test area and one in the control line indicate negative results. A negative result does not indicate the absence of amphetamines in the specimen . It only indicates that the level of amphetamines in the specimen is less than the threshold level .
One colored band appears in the control line . No colored band is found in the test line . This is an indication that the level of amphetamines in urine specimen is above the permitted limit. Invalid : If no colored band in control line zone - the test result is invalid. Retest the sample with a new test device.
Note : Very lightly colored band in the test line indicates that the amount of amphetamines in the sample is close to the level of permissible limit.
These specimens and other positive samples should be confirmed by an alternative method izpolzaveneto by GC / MS.

EXPECTED RESULTS
Amphetamine test is a qualitative analysis. It identifies the d-amphetamine in human urine in a concentration of 1000 ng / ml or higher. The concentration of d-amphetamine could not be determined by this assay . The test is designed to distinguish negative result of the alleged positive result. All positive results must be confirmed using an alternate method, preferably GC / MS
